Divarasib cut progression and death risks in lung cancer trial
Genentech tested divarasib in a Phase III study of people with lung cancer linked to a KRAS G12C mutation who had already received treatment.
The trial, called Krascendo 1, compared the drug with first-generation KRAS G12C inhibitors, including sotorasib and adagrasib.
Divarasib lowered the risk of disease progression or death by 42% compared with those inhibitors.
The overall risk of death was 44% lower with divarasib, and median overall survival was about 18 months, versus 12 months with the other drugs.
No new safety signals were observed, and the most common treatment-related side effects were mainly low grade and manageable.
The results were presented at the 2026 ESMO Congress and published in The New England Journal of Medicine; no further next steps were stated.
Divarasib reduced the risk of disease progression or death by 42% and the risk of death by 44% in a Phase III trial of previously treated KRAS G12C-mutated lung cancer.
Median overall survival was 17.7 months with divarasib, compared with 12 months with sotorasib or adagrasib.
The Krascendo 1 study compared divarasib alone with the two existing drugs in 338 patients.
Tumors shrank in 58% of patients receiving divarasib, compared with 32% in the comparison group.
Divarasib is not yet approved, and Roche said it is working with health authorities to make the drug available.
- Who
