Reddit analysis finds varied symptoms reported by users of unapproved retatrutide
Penn researchers used AI to study nearly 150,000 Reddit posts and comments about people who said they were taking retatrutide.
Retatrutide is an experimental weight-loss drug that acts on three hormone receptors and is still in Phase 3 trials, the final stage of testing before possible FDA approval.
The products discussed online are unapproved and are not intended for human consumption.
Among reports the researchers could classify, increased appetite was the most frequently reported symptom after expected effects such as weight loss and appetite suppression were excluded.
Compared with Reddit posts about approved drugs semaglutide and tirzepatide, retatrutide posts included proportionally more reports of sleep problems, faster heart rate and muscle changes, and fewer reports of nausea and other digestive symptoms.
The posts cannot prove retatrutide caused these symptoms or confirm that users took genuine retatrutide, because the products vary and their contents cannot be verified.
Further research is needed to understand what the reported differences mean as the drug continues through clinical testing.
Researchers at the University of Pennsylvania used AI to analyze nearly 150,000 Reddit posts and comments about unapproved retatrutide.
Among classifiable reports, increased appetite was the most frequently reported symptom after expected effects were excluded.
Compared with Reddit discussions of semaglutide and tirzepatide, retatrutide posts proportionally included more appetite-related symptoms, sleep disturbances, increased heart rate and muscle-related changes.
Nausea and other gastrointestinal symptoms were proportionally less common in the retatrutide posts.
The accounts cannot show that retatrutide caused the symptoms or confirm that users took genuine retatrutide.

This story does not have two clearly opposing sides.
Trials demonstrated weight-loss effects and identified nausea and other gastrointestinal complaints as the most common side effects.
Retatrutide is in the final stage of clinical testing before possible FDA approval.

What people are saying
We can’t say that retatrutide is causing these symptoms, or even that Reddit users are actually taking genuine retatrutide, since the drug is not legally available, and gray market formulations vary.
Retatrutide works differently from existing GLP-1 drugs, so it’s possible that its effects on the body are different, too.
Users willing to obtain an unapproved medication on the gray market likely differ from other clinical populations.
